Gel filtration column chromatography is suitable for multiple molecular size-based separations. Gel filtration columns can assist with the purification of proteins, lipoproteins, peptides, and other biomolecules. Processes such as desalting columns and buffer exchanges use gel filtration chromatography or size exclusion chromatography. These two processes are the most widely used gel filtration chromatography techniques, and they can be performed using the same materials. Gel filtration using columns carries the advantage of speed, and results can be achieved within a few minutes.

What is gel filtration used for?

Gel filtration, known as size-exclusion chromatography, separates molecules based on size. It is commonly employed in biochemistry and molecular biology to purify proteins, nucleic acids, or other macromolecules. The technique involves passing a mixture through a column packed with porous beads. Larger molecules move faster because they cannot enter the bead’s pores, while smaller molecules take longer as they diffuse into the pores.
