LB Broth (Miller formulation) is used for the growth and maintenance of E. coli containing recombinant plasmids. This media is nutritionally rich, which allows for rapid growth.
The Miller formulation is more commonly used than Lennox and Luria formulations, which both contain less NaCl than the Miller formulation. [Lennox formulation (0.5% NaCl) is often used to cultivate E. coli in the presences of salt-sensitive antibiotics (e.g., blasticidin, puromycin, and zeocin). Luria formulation (0.05% NaCl) can be used to optimize growth conditions by supplementing with additional NaCl.]
Powdered broth provides an economical option that requires less storage space and has a longer shelf life than liquid broth. Pre-weighed dry broth is packaged into foil pouches to make 500 or 1000 mL of LB broth. Alternatively, powdered LB broth in g to kg amounts provides the flexibility to make what you need.
