Application: Trypsin solutions, especially combined with EDTA, are used to release adherent cells from culture surfaces. Trypsin:EDTA solutions are generally formulated with natural porcine pancreas-derived trypsin, which generally also contain traces of chymotrypsin and elastase activity. Trypsin solutions can range from 0.025% to 0.5%. The reasons for the range of concentrations are differences in required trypsin activity or potency; differences in incubation times; and cell line susceptibility. The concentration of trypsin necessary to dislodge cells from their substrate is dependent primarily on the cell type and the age of the culture. Various formulations should be developed and tested to determine the best product for a specific application.

Biochem/physiol Actions: Tyrpsin cleaves peptide chains mainly at the carboxyl side of the amino acids lysine or arginine. Tyrpsin plays an important role in digestion and protein absorption. Tyrpsin originates in the pancreas in the form of the zymogen, trypsinogen1.
