Neurofilaments are composed of three subunits, namely NEFL (light), NEFM (medium) and NEFH (heavy). NEFH (neurofilament heavy) gene encodes the most important neuron-specific intermediate filament of cytoskeleton in myelinated axons. Biochem Physiol Actions: The heavy polypeptide neurofilament encoded by neurofilament heavy chain (NEFH) gene participates in the maintenance of cytoskeleton and axonal architecture in the proximal axonal region of spinal motoneurons. Mutations in the NEFH gene has a key role in the pathogenesis of sporadic amyotrophic lateral sclerosis (ALS). NEFH gene is also identified to generate autosomal dominant axonal Charcot-Marie-tooth disease (CMT2cc).
