Components: Sigma's Lambda DNA Mixed Digest is provided in a solution of 10 mM Tris-HCl (pH 8.0), with 1.0 mM EDTA.
Preparation Note: Bacteriophage lambda DNA was completely digested with by Apa I, Kpn I, Xba I and Xho I in separate reactions, sticky ends filled with DNA polymerase I Klenow (to prevent reannealing), mixed with uncut bacteriophage lambda DNA, and then purified by phenol extraction.
Other Notes: For optimal resolution, the recommended agarose gel concentration is 0.4%.
