IPTG, Isopropyl ß-D-1-thiogalactopyranoside, is a non-metabolizable galactose analog used to induce cells to produce a protein that the lac operator controls. It functions by binding to the lacI repressor and altering its conformation, which prevents the repression of genes controlled by the lac operator.
IPTG is commonly used in cloning procedures that require induction of ß-galactosidase activity. It induces expression of the lac operon in Escherichia coli, which includes lacZ that codes for ß-galactosidase. X-Gal or Bluo-Gal can be used in this procedure to perform blue-white selection of recombinant bacterial colonies.
