Biochem Physiol Actions: HAMNO is a potent and selective inhibitor of replication protein A (RPA) interactions with proteins involved in the replication stress response. HAMNO binds the N-terminal domain of RPA70 and inhibits both ATR autophosphorylation and phosphorylation of RPA32 Ser33 by ATR. HAMNO enhances DNA replication stress in cancer cells and slow tumor growth in vivo.
