Yes-associated protein 1 (YAP1) is a transcriptional coactivator and has two WW domains (two conserved tryptophans), TID domain (TEA domain family member 1 (TEAD) transcription factor interacting domain), proline rich region, sarcoma homology 3 domain (SH3) binding motif and post synaptic density protein (PSD95), Drosophila disc large tumor suppressor (Dlg1), and zonula occludens-1 protein motif (PDZ). It is expressed in eight isoforms. The gene YAP1 is mapped to human chromosome 11q22.1.

Biochem Physiol Actions: Yes-associated protein 1 (YAP1) interacts with the src family kinase c-Yes by its SH3 binding motif and with protein 73 using WW motif. YAP promotes cell and tissue growth. YAP1 interacts with Receptor tyrosine-protein kinase (ErbB4) receptor and regulates transcription. YAP is oncogenic and contributes to tumor progression. Phosphorylation of YAP is critical for regulating apoptosis. YAP1 levels are elevated in human cancers. It is highly expressed in hedgehog-associated medulloblastomas. Mutations in YAP1 is also implicated in optic fissure closure defects. Knockdown of YAP1 gene impacts on prostate cancer progression and is considered as potential target for treatment.
