Vacuolar protein sorting-associated protein 4B (EC 3.6.4.6; UniProt O75351; also known as Cell migration-inducing gene 1 protein, Protein SKD1, Suppressor of K(+) transport growth defect 1) is encoded by the VPS4B (also known as SKD1, SKD1B, VPS4-2, VPS42, MIG1) gene (Gene ID 9525) in human. SKD1 is a ubiquitously expressed protein that is involved in intracellular protein trafficking at the late endosomal protein transport step. Displays about 80% sequence homology with VPS4A. It recognizes membrane-associated ESCRT-III (endosomal sorting complex required for transport III) assemblies and catalyzes their disassembly, possibly in combination with membrane fission. Also, redistributes the ESCRT-III components to the cytoplasm for further rounds of multi-vesicular body (MVB) sorting. In conjunction with the ESCRT machinery, it also appears to function in topologically equivalent membrane fission events, such as the terminal stages of cytokinesis and enveloped virus budding (HIV-1 and other lentiviruses). VPS4A/B are required for the exosomal release of SDCBP, CD63 and syndecan. SKD1 is proposed to be monomeric or homodimeric in nucleotide-free form and to oligomerize upon binding to ATP to form two stacked hexameric or heptameric rings with a central pore through which ESCRT-III substrates are translocated in an ATP-dependent manner.Ref.: Scheuring, S et al. Application: Immunocytochemistry Analysis: A representative lot detected VPS4B immunoreactivity concentrated at the centrosome during mitosis and at the midbody during cytokinesis in 3% paraformaldehyde-fixed HeLa cells. No staining was seen in VPS4B shRNA-transfected HeLa cells (Morita, E., et al. (2010). Proc. Natl. Acad. Sci. U.S.A. 107(29):12889-12894).Western Blotting Analysis: A representative lot detected the VPS4B target band in lysate from untransfected, but not VPS4B shRNA-transfected HeLa cells (Morita, E., et al. (2010). Proc. Natl. Acad. Sci. U.S.A. 107(29):12889-12894).
