USP14 is made up of 494 amino acids containing a 45kDa catalytic domain at its N-terminus. It contains a ubiquitin-like (Ubl) domain at the N-terminus that associates with 19S regulatory particle (RP) 26S proteasome. This binding activates the deubiquitinating activity of the protein.

Biochem Physiol Actions: USP14 (ubiquitin specific peptidase 14) gene encodes a protein that belongs to the ubiquitin-specific processing (UBP) family of proteases. It is a proteasome-associated deubiquitinating enzyme (DUB) containing His and Cys catalytic domains. It releases ubiquitin from the proteasome targeted ubiquitinated proteins resulting in regeneration of ubiquitin at the proteasome. It is involved in the degradation of chemokine receptor CXCR4. CXCR4 is required for CXCL12-induced cell chemotaxis and its overexpression leads to cancer and inflammatory and neurodegenerative disorders. Under ER stress, It binds to a, a endoplasmic reticulum (ER) stress receptor, that is involved in unfolded protein response (UPR), a mechanism to control the accumulation of unfolded proteins within the (ER) lumen. Its overexpression leads to ER-associated degradation (ERAD) pathway inhibition.
