Probable helicase senataxin (UniProt Q7Z333; also known as Amyotrophic lateral sclerosis 4 protein, SEN1 homolog) is encoded by the SETX (also known as ALS4, AOA2, SCAR1, bA479K20.2) gene (Gene ID 23064) in human. Senataxin is implicated in both genome and RNA transcriptome homeostasis. Senataxin contains a helicase domain for mediating RNA interaction and an N-terminal domain for critical protein interactions. Senataxin is reported to colocalize with DNA repair proteins, including the DNA damage response protein 53BP1, at distinct nuclear foci in S/G2-phase human cells, and the number of these foci increases in response to impaired DNA replication or DNA damage. Senataxin is shown to resolve RNA/DNA hybrids (R loops) that arise during transcription, which can lead to genome instability unless they are resolved efficiently.
