The gene S100A8 (S100 calcium-binding protein A8) is mapped to human chromosome 1q21. It is a small calcium-binding protein, which is strongly expressed in neutrophils. In presence of cell stimulation, it is also expressed in monocytes, macrophages, platelets and epithelial and endothelial cells. S100A8 protein is present as a homodimer as well as heterodimer (S100A8/A9). The protein has two EF-hand motifs and one high-affinity Ca2+ binding site. S100A8 is also referred to as myeloid-related protein 8 (MRP8) or calgranulin A.

Biochem Physiol Actions: S100A8 (S100 calcium-binding protein A8) is mainly present at the inflammation site or in the serum during acute and chronic inflammation. It is responsible for neutrophil recruitment, attachment and release from the bone marrow. S100A8 is required for sending neutrophils to the site of inflammation in presence of bacterial infection, lipopolysaccharide, and monosodium urate crystals. It is also involved in granulocyte and monocyte attachment to endothelium and also their transport across endothelial cells. The S100A8/A9 heterodimer induces monocyte migration and cytokine secretion. In presence of falciparum malaria, high levels of S100A8 are present in the blood.
