Biochem Physiol Actions: RAD21 (RAD21 homolog, S. pombe) plays a crucial role in normal cell division. It acts as a main component of the cohesin complex for the proper chromosomal arrangement during cell division. The protein possesses two mitotic cleavage sites for the attachment of separase. It has been shown experimentally that at the beginning of anaphase stage, a caspase-like Esp1/separase attaches to the cleavage site of RAD21 and initiate sister chromatid separation, which is an essential step for the completion of cytokinesis. During double strand DNA damage repair, it helps the two DNA strands to adhere with each other. Mutation in RAD21 causes a congenital phenotype, cohesinopathy in humans characterized with growth retardation and minor skeletal anomalies.
