Biochem Physiol Actions: PTPN22 (protein tyrosine phosphatase, non-receptor type 22) gene encodes a member of the non-receptor class 4 subfamily belonging to the protein-tyrosine phosphatase family. The encoded protein is a lymphoid-specific intracellular phosphatase that negatively regulates T-cell receptor (TCR) signaling. It dephosphorylates the Src family kinases and certain other key signaling molecules involved in TCR signaling. It functions in the development and activation of lymphocytes, and in innate immune cell-mediated host defense and immunoregulation. The gene is localized to human chromosome 1p13, a region associated with the incidence of type 1 diabetes in certain populations. Mutations in this gene are linked to increased susceptibility to autoimmunity and infection.
