Biochem Physiol Actions: Progesterone receptor (PGR) functions as a transcription factor which modulates the transcription of target genes in response to progesterone and other hormones. It is phosphorylated on multiple sites and at times the phosphorylation changes according to the hormone involved. In the absence of hormones, PGR is complexed with heat shock proteins. When the ligand binds, it causes the release of PGR from heat shock proteins. It is then translocated to the nucleus where it binds to its DNA response elements (SREs) and to the components of the transcription machinery. PGR also binds to chromatin templates and results in chromatin remodeling adjacent to the PGR-binding sites.
