PGAM5 is a mitochondrial specific phosphatase specific for serine/threonine residues. PGAM5 principally dephosphorylates and activates MAP3K5 kinase a critical player in many cancers. PGAM5 also acts as a central mediator for programmed necrosis or necroptosis, induced by TNF, ROS or calcium. In the mitochondria, PGAM5 may also function as a regulator of mitochondrial dynamics and reconstruction. Also PGAM5 functions in the cellular stress response and regulates the removal of damaged mitochondria via mitophagy. PGAM5 itself is regulated at the protein level by the mitochondrial specific rhomboid protease PARL, which cleaves the N-terminal of PGAM5 in response to mitochondrial depolarization.
