PARP4 (Poly ADP-ribose polymerase family, member 4) is a 193kDa nuclear, NAD-dependent, vault protein encoding an ADP-ribosyltransferase-like protein. It is a subunit of ribonucleoprotein (RNP) complex belonging to the inter-a-inhibitor family. Structurally, it is composed of a catalytic domain similar to the nuclear-localized ADP-ribosyltransferase proteins (Adprt), two Adprt-like proteins (Adprtl2 and Adprtl3), and the telomere-associated protein tankyrase.

In DNA assembly, it crucially identifies the DNA nick sites and rejoins the DNA strands. It poly(ADP-ribosyl)ates the nuclear proteins in response to any DNA damage.
