Kinetochore protein Nuf2 (UniProt Q9BZD4; also known as Cell division cycle-associated protein 1, hNuf2, hNuf2R, hsNuf2) is encoded by the NUF2 (also known as CDCA1, NUF2R) gene (Gene ID 83540) in human. Nuf2 is a kinetochore complex component that plays an important role in stabilizing microtubule attachment. Nuf2 knockdown results in kinetochore attachment defects and eventual death of mitotic cells. Nuf2 binds and stabilizes CENP-E in the kinetochore, and both proteins are essential for proper chromosome segregation during mitosis. Nuf2 is also shown to complex with Hec1, and the Nuf2-Hec1 complex is essential for organizing microtubule attachment sites. Nuf2 upregulation is reported in various cancers, and Nuf2 knockdown results in significant cell growth delay and apoptosis among non-small cell lung, colorectal, gastric, and ovarian cancer cell lines.
