NNT (nicotinamide nucleotide transhydrogenase) is an integral protein of the inner mitochondrial membrane. It is composed of three subunits termed dI, dII and dIII. The dI component binds to the NAD(+)/NADH, and the dIII binds to the NADP(+)/NADPH, protrude from the membrane.

Biochem Physiol Actions: NNT (nicotinamide nucleotide transhydrogenase) acts as an antioxidant defense gene. It catalyzes the transhydrogenation reaction of transferring hydride ion from NAD(H) to NADP(H). In reverse transhydrogenation, it acts by the outward proton translocation and formation of a proton motive force. It has been reported that NNT may also participate in reactive oxygen species (ROS) detoxification in human adrenal glands. Mutations in NNT gene cause familial glucocorticoid deficiency.
