MAFA is a tissue specific transcription factor found in the nuclei of pancreas islet beta cells. MAFA specifically binds the insulin enhancer element RIPE3b and activates insulin gene expression. MAFA also cooperates synergistically with NEUROD1 and PDX1 in enhancing additional gene expression during development as well. Phosphorylation by GSK3 increases MAFA transcriptional activity and is required for its oncogenic activity. MAFA can be either an oncogene or act as a tumor suppressor, depending on the cell context.
