Low-density lipoprotein receptor-related protein 2 (UniProt P98164; also known as Calcium sensor protein, Glycoprotein 330, gp330, Heymann nephritis antigen homolog, LRP-2, Megalin) is encoded by the LRP2 (also known as DBS) gene (Gene ID 4036) in human. Megalin/LRP-2 is a 600 kDa type I membrane receptor initially produced with a signal peptide sequence (a.a. 1-25). The mature receptor has a large extracellular region (a.a. 26-4423), a transmembrane segment (a.a. 4424-4446), and cytoplasmic tail (a.a. 4447-4655). Megalin belongs to the LDLR family of membrane glycoproteins whose members also inculde the low-density lipoprotein receptor (LDLR), the very-low-density lipoprotein (VLDL) receptor, the LDLR-related protein (LRP), and the apolipoprotein E receptor 2 (ApoER2). The overall extracellular structural features are similar among all LDLR family members. Megalin contains four clusters of cysteine-rich Complement-type/LDLR Class A repeats that mediate ligand-binding. These are separated by 17 EGF-like repeats and eight cysteine-poor spacer regions that contain YWTD motifs involved in the pH-dependent ligands dissociation in endosomal compartments. Megalin is a multiligand, endocytic receptor expressed primarily in the apical membranes of polarized epithelial cells. In addtion to functioning alone, megalin is also reported to interact with another structurally distinct endocytic receptor cubilin in several tissues where they are co-expressed. The internalization of several cubilin ligands is reported to be strongly facilitated in the presence of megalin.
