The pro-interleukin-16 (IL16) gene is mapped to human chromosome 15q26.3. IL16 mRNA is present particularly in leukocytes. It is produced as a large precursor protein which is cleaved by caspase-3 and released as a carboxyl-terminal 121-amino acid fragment. It was formerly called as lymphocyte chemoattractant factor.

Biochem Physiol Actions: Pro-interleukin-16 (IL16) is a multifunctional cytokine with roles in inflammation and autoimmune diseases. IL16 binds CD4 receptor and can activate CD4+ T cells, monocytes, macrophages, eosinophils and dendritic cells. Also, it stimulates monocyte to secrete tumor-associated inflammatory cytokines tumor necrosis factor-a, IL-1ß, IL-6 and IL-15 which are associated with tumorigenesis. During human T cell leukemia virus type-1 (HTLV-1) infection, the HTLV-1 TAX oncoprotein binds precursor IL16 and causes G1/S phase progression in lymphocytes. Single nucleotide polymorphism in IL16 is associated with increased risk of glioma, non-cardia gastric cancer, nasopharyngeal carcinoma and colorectal cancer.
