IFI16 (?-interferon-inducible protein 16) is a member of the PYRIN superfamily and HIN-200 family (hematopoietic interferon-inducible nuclear antigens with 200 amino acid repeat) which is involved in the p53-mediated regulation of apoptosis and inflammation signaling pathways. It is highly expressed in endothelial cells and squamous stratified epithelia. It is found in the nuclei of lymphocytes in the spleen, thymus, lymph nodes, palatine tonsil and non-lymphoid tissues including trachea, gastrointestinal tract, skin and testis. IFI16 is composed of a DNA binding domain, a transcriptional regulatory domain, DAPIN (domain in apoptosis and interferon response) domain associated with interferon (IFN) response and BRCA1 binding domain (breast cancer tumor suppressor protein).

Biochem Physiol Actions: IFI16 (?-interferon-inducible protein 16) is involved in the regulation of p53-mediated gene transcription. It acts as a potent transcription repressor. It stimulates p53-mediated apoptosis under DNA damage conditions. Along with p53, it also regulates other target genes such as p21, HDM2 (E3 ubiquitin-protein ligase), and BAX (apoptosis regulator) in MCF7 cells.
