Biochem Physiol Actions: The gene ENTPD1 (ectonucleoside triphosphate diphosphohydrolase 1) encodes a plasma membrane-bound vascular ATP diphosphohydrolase (ATPDase) that catalyzes the hydrolysis of extracellular ATP and ADP to adenosine, which associates with adenosine receptors and negatively regulates T-cell and natural killer (NK)-cell responses, resulting in a suppressed immune system. The regulatory T cell (Treg) immunosuppressive function is mainly due to the CD39/CD73 pathway that produces adenosine. The expression of CD39 is found to be higher in cancer cells than in normal cells. It is expressed by infiltrating lymphocytes, the tumor stroma, and tumor cells in cancer tissues. It may serve as a therapeutic target for inhibiting tumor cell-mediated immunosuppression.
