EMX1 (empty spiracles homeobox 1) gene is localized to human chromosome 2p14-p13. It is a late and narrow expression gene. It is a homeodomain transcription factor. It is a human ortholog of ems (empty spiracles) protein found in Drosophila. This protein is a resident of nucleus. In embryos, the expression of this gene is restricted to cerebral cortex and olfactory bulbs.

Biochem Physiol Actions: EMX1 (empty spiracles homeobox 1) interacts with WDR11, which is a part of olfactory neuron development. In developing central nervous system, this protein determines cell fates. It participates in sonic hedgehog-Patched-Gli (Shh-Ptch-Gli) signaling pathway, where it acts as an effector protein for Gli-Kruppel family member 3. EMX1-lineage derived cells are responsible for two types of neuronal population in mature basal forebrain namely, a subclass of distinct amygdala excitatory neurons, and medium spiny neurons (MSNs) in the striatum. Thus, this protein determines the striatum and amygdala fate of developing neurons.
