Biochem Physiol Actions: EIF2AK2 (Eukaryotic translation initiation factor 2-alpha kinase 2) is a serine/threonine protein kinase involved in phosphorylation of translation initiation factor EIF2S1 and in turn EIF2AK2 mediated phosphorylation directly restricts protein synthesis. The activity of EIF2AK2 is activated by autophosphorylation after binding to dsRNA, manganese ions and heparin. It also participates in the regulation of apoptosis in response to ER stress. It has also been reported that EIF2AK2 can be a potential target for neuroprotective therapeutics. It is involved in the activation of NF-?B pathway by interacting with IKBKB (I?B kinase ß) and TRAF (Tumor necrosis factor receptor-associated factor) family of proteins. EIF2AK2 also activates p38 MAP kinase pathway upon interacting with MAP2K6. It plays a key role in the regulation of the innate immune response.
