DnaJ homolog subfamily B member 1 (UniProt Q3MI00) is encoded by the DNAJB1 gene (Gene ID 538426) in bovine species. Heat shock proteins (Hsps) are classified into six major families based on their relative molecular mass, Hsp110/hspH, Hsp90/HspC, Hsp70/HspA, Hsp60/HspD, Hsp40/DnaJ, and small heat shock proteins sHsp/HspB. Hsp40/DnaJB1 belongs to the Hsp40/DnaJ subfamily of heat shock proteins. Hsp40 acts as a co-chaperone with Hsp70 to facilitate protein folding, transport, and degradation. MK5 is reported to stimulate the ATP hydrolyse activity of Hsp40/Hsp70 complex by phosphorylating Hsp40/DnaJB1 C-terminal serine residues Ser149, Ser151, Ser171, resulting in an enhancement of Hsp40/DnaJB1-dependent suppression of heat shock factor 1-driven transcription. DnaJB1 is also shown to interact with MDM2 and prevent MDM2-mediated ubiquitination and proteasomal degradation of p53. DnaJB1 depletion promotes cancer cell growth as a result of p53 pathway inactivation.
