Biochem Physiol Actions: CSF1R (Colony stimulating factor 1 receptor) is a major molecule in the innate immunity, cancer, and inflammatory diseases, including systemic lupus erythematosus, arthritis, atherosclerosis and obesity. It regulates developmental activities including cell survival, proliferation, differentiation, and function of mononuclear phagocytes. It has been reported that CSF-1 autocrine loop helps to activate macrophages. In actual state, it exists as an autoinhibited form, and upon activation, it dimerizes. Later, it autophosphorylates tyrosine residues in the intracellular domain, followed by the recruitment of signaling molecules as well as internalization of the receptor. Mutation in the CSF1R causes several diseases including myeloid malignancies.
