CD276 (cluster of differentiation 276) is a member of the B7 family of T cell co-regulatory molecules, which functions as an immune-regulatory molecule. This protein is inducible by inflammatory cytokines on T-cells, B-cells and dendritic cells. Its ligand is yet unidentified.

Biochem Physiol Actions: CD276 (cluster of differentiation 276) functions as both T-cell stimulatory and inhibitory protein in tumor cells, and hence, its function in tumors is not very clear. In CRC (colorectal cancer), the expression of this protein has a strong and independent relation with poor prognosis. The expression of this protein is highly increased in endothelial cells (ECs) in cancer as opposed to that in normal tissue. Also, the number of circulating ECs is increased in tumor conditions as compared to normal conditions. CD276 is also up-regulated in glioblastomas, breast, lung, renal and colon carcinomas. It is a putative biomarker for rheumatoid arthritis (RA), and B7-H3-T-A-C-T polymorphism is linked to increased RA risk. This variant might also be linked to the secretion of soluble B7-H3. CD276 promotes tumorigenesis of mantle cell lymphoma, and its inactivation leads to increased sensitivity to chemotherapy.
