Chlortetracycline Hydrochloride

Chlortetracycline hydrochloride has antimicrobial action similar to tetracycline hydrochloride, but it is somewhat less active against many Gram-negative organisms. It was first isolated from the culture of Streptomyces aureofacien.

Chlortetracycline is an antibiotic produced by some strains of Streptomyces aureofaciens. Mode of Action: Inhibits protein synthesis (elongation) by preventing binding of aminoacyl-tRNA to the 30S subunit. Antimicrobial spectrum: Gram-positive. Less active against Gram-negative bacterial than tetracycline. Mode of Resistance: Loss of cell wall permeability.

To prepare Stock Solutions: Add the appropriate amount of powder to water while mixing (not the reverse), as adding water to the powders may result in insoluble mixtures that cannot be filtered without losing potency. If desired, the pH may be adjusted, but a basal media with greater than 2 g/L sodium bicarbonate can easily absorb up to 2 % of an acidic antibiotic solution; therefore, an antibiotic solution with an acidic pH would not influence the pH of the media. After ensuring that the resultant solution is transparent, sterile filter and store in small aliquots at appropriate temperature until use. Prior to use of the media, aseptically add the appropriate amount of antibiotic to the basal media in order to obtain the required working concentration. Mix well and sterile filter if asepsis is believed to have been compromised.

Solubility: The product is tested in 1 M NaOH, giving a clear yellow to brown solution at 50 mg/mL. Its solubility at room temperature in water is reportedly about 8.6 mg/mL, in methanol, 17.4 mg/mL and in ethanol, 1.7 mg/mL. It is soluble in solutions of alkali hydroxides and carbonates. It is practically insoluble in acetone and other organic solvents.(Lit.)

Sterilization of Solutions: Solutions should be filter sterilized. Store in small aliquots at appropriate temperature until use.
