The Soil DNA Extraction Kit was designed for raid isolation of genomic DNA from microorganisms such as bacteria, archaea, fungi, and algae in soil samples. The soil sample is homogenized and disrupted using SL1 (lysis) Buffer combined with ceramic beads. Insoluble particles, proteins and PCR inhibitors such as humic acid are then precipitated with a unique inhibitor removal buffer (SL2). In addition, residual PCR inhibitors remaining in the clear supernatant are further removed by passing through a specialized PCR Inhibitor Removal Column. The flow-through is then mixed with a binding buffer (SL3) and the genomic DNA is bound by the GD Column. The column is then washed and the DNA is eluted with Elution Buffer. DNA phenol extraction or alcohol precipitation is not required and the entire procedure can be completed within 40 minutes. The purified genomic DNA is ready for use in PCR, restriction enzyme digestion, and sequencing reactions.
The quality of the Soil DNA Extraction Kit is tested on a lot-to-lot basis by isolating genomic DNA from 250 mg soil samples.
