All human products should be treated as potentially infectious. Source material from which this product was derived was found negative when tested in accordance with current FDA required tests. No known test methods can offer assurance that products derived from human blood will not transmit infectious agents. The total protein is determined using the Biuret procedure, with bovine albumin as the standard. Each vial is overfilled by approximately 5 percent so that a minimum of 5 mg of human secretory IgA is obtained upon reconstitution.
Human secretory IgA is purified from pooled human colostrum using multi-step procedures which may include salt fractionation, gel filtration, ion-exchange chromatography and immunoabsorption. The product is dialyzed into 0.01M sodium phosphate, 0.07M sodium chloride, pH 7.3, filtered through a 0.22 µm filter, vialed and lyophilized.
IgA secretory antibody is present abundantly in mucous linings of gastrointestinal, respiratory and genitourinary tracts, tears, colostrum and saliva. Thus secretory IgA contributes to the humoral defense mechanism against the pathogens on mucosal surfaces.
Purified IgA secretory may be used as a reference antigen, standard, blocking agent or coating protein in a variety of immunoassays including ELISA, dot immunobinding, immunoblotting, immunodiffusion and immunoelectrophoresis. Other applications include starting materials for the preparation of immunogens and solidphase immunoadsorbents. IgA from human colostrum was used as a standard in ELISA (1 mg/mL) and gel electrophoresis.
Product is the lyophilized powder of human secretory IgA and buffer salts.
Reconstitution: Reconstitute product with 1 mL of deionized or distilled water. Gentle swirling may be used to speed rehydration. Avoid vigorous shaking of the reconstituted material.
Assay: The product is tested for purity at a minimum of 20 mg/mL using immunoelectrophoresis. The product is human secretory IgA; no trace of IgG, IgM or non-immunoglobulin serum proteins is detected.
