The product is tested for purity at a minimum of 20 mg/mL using immunoelectrophoresis. The product is human IgM; no trace of IgG, IgA or non-immunoglobulin serum proteins is detected. The total protein is determined using the Biuret procedure, with bovine albumin as the standard. Each vial is overfilled by approximately 10 percent so that a minimum of 5 mg of human IgM is recovered.
Human IgM is purified from human myeloma serum using multi-step procedures which may include salt fractionation, gel filtration, ion-exchange chromatography and immunoabsorption. The product is dialyzed into 0.01M sodium phosphate, 0.07M sodium chloride, pH 7.3, filtered through a 0.22 μm filter and vialed. No preservative has been added.
Purified human IgM is suitable for use as a positive control in antibody identification screening techniques and is recommended as an ELISA coating antigen and control.
