SmartGlow™ Pre Stain can be used the same way as Ethidium Bromide for agarose and polyacrylamide gel electrophoresis. The stain emits green fluorescence when bound to dsDNA or ssDNA and emits red fluorescence when bound to RNA. SmartGlow™ stain has two excitation peaks at approximately 290nm (UV light) and 490nm (Blue light).

Protocol:

  1. Prepare 100ml of agarose or polyacrylamide solution, heat to boil.
  2. Add 5ul of SmartGlow™ stain to the gel solution.
  3. Mix gently to and make sure there are no air bubbles in the mixture
  4. Allow the solution to cool to about 60°C to 70°C then pour the gel(s).
  5. For enhanced results, you can optionally add 5ul of SmartGlow™ stain per 100ml of the running buffer. Adding the stain to the running buffer can help in increasing sensitivity and detecting small quantities of nucleic acid.
  6. After the separation is complete, view the gel using a UV or blue light illuminator.

Using SmartGlow™ Loading Dye, it is not required to add any stain to the gel or running buffer. SmartGlow™ Loading Dye is supplied at 6X concentration, and only needs to be added to the samples before pipetting into the gel wells. This method eliminates the possibility of contamination of glassware, plasticware, and the gel running tank as can happen with EtBr. After electrophoretic separation, view and document your results using a UV or Blue Light Illuminator.

Protocol:

  1. Prepare 100ml of agarose or polyacrylamide solution

  3. Allow the solution to cool to about 60°C to 70°C then pour the gel(s).
  4. Mix samples and DNA markers with SmartGlow™ Loading Dye at a 1:5 (dye to sample) dilution.
  5. After the separation is complete, view the gel using a UV or blue light illuminator.

SmartGlow™ products are considered safer than Ethidium Bromide. They are non-carcinogenic as determined by the Ames-test, with negative results in both mouse marrow chromophilous erythrocyte micronucleus and mouse spermary spermatocycte chromosomal aberration tests.

    I was able to try the SmartGlow PS stain on my post PCR gels - quite good results - easy to use. You mix a little bit with the gel mixture just like using EtBR. Used a UV trans to view the gels. Thomas was nice enough to send me a sample so I could make sure it worked well before buying.
