LB Agar Plates (Miller formulation) are used for the growth and maintenance of E. coli containing recombinant plasmids. This media is nutritionally rich, which allows for rapid growth.
The Miller formulation is more commonly used than Lennox and Luria formulations, which both contain less sodium chloride than the Miller formulation. [Lennox formulation (0.5% sodium chloride) is often used to cultivate E. coli in the presences of salt-sensitive antibiotics (e.g., blasticidin, puromycin, and zeocin). Luria formulation (0.05% sodium chloride) can be used to optimize growth conditions by supplementing with additional sodium chloride.]
Typically, 100 mm round plates are for growth/selection, 150 mm round plates are for screening, and rectangular plates can be used with automated systems.
Contains 1.0% (10 g/L) tryptone, 0.5% (5 g/L) yeast extract, 1.0% (10 g/L) sodium chloride, and 1.5% agar.
