Ready-to-use 2X YT Agar Plates are used for growth of recombinant E. coli and propagation of M13 bacteriophage in E. coli. This highly nutritious medium encourages rapid growth that is further enhanced by the addition of the carbon source glucose. Glucose is also helpful for growing fastidious organisms and low inocula and reviving older cultures.
Carbenicillin is used to select for mutated or transformed cells that carry the ß-lactamase (bla) gene, which confers resistance to ampicillin (Amp or AmpR) and carbenicillin. Carbenicillin is a semi-synthetic ampicillin analog that does not allow formation of satellite colonies.
Typically, 100 mm plates round plates are for growth/selection, while 150 mm plates round plates are for screening. Rectangular plates can be used with automated systems.
Contains 1.6% (16 g/L) tryptone, 1.0% (10 g/L) yeast extract, 0.5% (5 g/L) sodium chloride, 2% glucose, 100 µg/ml carbenicillin, and 1.5% agar.
