Biochem Physiol Actions: H6PD (hexose-6-phosphate dehydrogenase, glucose 1-dehydrogenase) is a microsomal enzyme involved in the regulation of peripheral corticosteroid metabolism in the liver and adipose tissue. It regenerates NADPH within the endoplasmic reticulum via the pentose phosphate pathway which is important for steroid hormone and drug metabolism. By supplying NADPH, it regulates the oxo-reductase activity of, 11-ß hydroxysteroid dehydrogenase type I (11ß-HSD1). The H6PD generated NADPH helps in reduction reaction of cortisone to cortisol in association with 11ß-HSD1. Mutation in H6PD leads to the polycystic ovary syndrome (PCOS) and obesity with hyperandrogenemia.
