The total protein is determined using the Biuret procedure, with bovine albumin as the standard. The product is tested for purity at a minimum of 20 mg/mL using immunoelectrophoresis. The IgG concentration is determined by radial immunodiffusion and is reported in mg/mL, and percentage of total protein.
Human IgG positive control is purified from pooled human serum using multi-step procedures which may include salt fractionation, gel filtration, ion-exchange chromatography and immunoabsorption. The product is dialyzed into 0.01M sodium phosphate, with 0.07M sodium chloride, pH 7.3, filtered through a 0.22 μm filter, vialed and lyophilized. No preservative has been added.
Immunoglobulin G (IgG) is one of the most abundant proteins in serum. IgG is important for our defence against microorganisms and the molecules are produced by B lymphocytes as a part of our adaptive immune response. The IgG molecule has two separate functions: to bind to the pathogen that elicited the response and to recruit other cells and molecules to destroy the antigen. The variability of the IgG pool is generated by somatic recombination and the number of specificities in an individual at a given time point is estimated to be 1011 variants.
Human IgG is suitable for use as a positive control in antibody identification screening techniques. Positive controls are not intended for use as calibration controls. Purified proteins are recommended as ELISA coating antigens and as controls.
Human IgG positive control is the lyophilized powder of human IgG positive control and buffer salts. IgG is a purified antigen.
