Tris-Glycine Buffer is used for Western blotting and gel electrophoresis. Tris-glycine buffer is used to make a Tris-glycine-methanol transfer buffer, which is the most common protein transfer buffer for wet blot transfers. The methanol prevents the gel from swelling during the transfer and enhances the protein binding to nitrocellulose.
Empty contents of 1 packet into a 4-liter flask, add 800 mL of methanol and QS to volume with deionized water. Four liters of pH 8.3 buffer is now ready to use.
