Mannitol Selenite broth w/ Brilliant green is recommended for enrichment of Salmonellae from faeces, food- stuffs and other materials. Sodium taurocholate, selenite, brilliant green inhibits gram positive bacteria and other gram negative bacteria. Mannitol is a fermentable carbohydrate and source of carbon. Do not incubate enriched culture longer than 24 hours as the inhibitory effect of selenite is reduced after 6-12 hours incubation. Further subculture the broth from the upper third of the broth column to greater or lesser inhibitory selective agars such as Salmonella Shigella Agar, MacConkey Agar, XLD agar etc.
